The following is based solely on my experience and involves sidequests directly connected to the Iron Clan quest. You could simply check the Arcanum Walthrough on this site, but I just like to test myself.
By the way, I'll highlight the entire text since it mostly consists of spoilers.
Spoiler
Your first encounter with the Iron Clan should be in Tarant. You'll find an Ancient Iron Chest in the museum holding Bellerogrim's skull replica and a dragon egg. At this point you won't be able to open it, unless you use up a fate point for a critical success in lockpick.
As you progress through the game, you'll eventually find yourself in the caverns of the Wheel Clan. There, in the residential area, you will locate a dwarf historian who will mention the Iron Clan to you (this is, by the way, the first time you're actually introduced to the existence of the Iron Clan) and ask you to find its location and retrieve the Durin stone from there for him. If you accept the quest, you'll get a schematic for the Iron Key. To create it, you'll need some mithril ore and a Heartstone. The former can be found in the Wheel Clan mines or purchased from a blacksmith there (for quite a high price, though). As for the Heartstone, you'll need to wait until you get to Qintarra.
When you get there, an elven hunter standing near the entrance to the upper level of the city will ask you to find his student who went missing. He will mark the Bedokaan village on your map, where you will find three heartstones total. Unless you're doing the Ancient Gods quest, you'll only need one to forge the Iron Key.
When you have the Iron Key, go back to Tarant and use it on the Iron Chest. In it you'll find three Iron Drums and a map to the Place of Lost Voices. At that location you'll find an ancient Iron Device that needs those three drums to operate. There is a sequence in which you need to insert them (which I can't recall of at the moment), otherwise you'll get attacked by several Mechanical Arachnids. After you insert all the drums, you'll get the message to find the Book of Durin's Truth.
Later on in the game you'll find yourself in Caladon, searching for Victor Misk. In his house you'll find a book located in a display case - the Book of Durin's Truth. You will be able to purchase it off Ms. Misk for 2000 gold. After you read it, the location of the Secret Entrance to the Iron Clan will appear on your map. However, you still won't be able to enter there for some time, because you'll need the Glass Key.
You'll have to finish the Tsen'Ang quest first. Only after you return from there, go back to Caladon and you'll find out that Alexander, the First Acolyte of the Panarii Temple has returned, giving you access to the inner parts of the temple. You'll need to speak to the two men in the rooms behind Alexander - Hadrian the historian and the other bloke whose name I forgot. :P
Anyway, you'll eventually get the quest to investigate Nasrudin's crypt. You can get to it through the sewers in front of the temple. When you get to the crypt, open the sarchopagus, examine it, grab the skull and then examine the lid. When you get back to the temple, show Hadrian the skull and eventually you'll find out that the person buried in there is, in fact, St. Mannox, and not Nasrudin. After that, talk to the other man and tell him about the sarchopagus lid. Go back to Alexander and inform him of everything you've discovered, and then offer your services of finding the truth about St. Mannox.
Now, you're off to Roseborough. The only crucial part of the Iron Clan quest here is located far to the north. Just keep walking along the coast and you'll run into a stone vault - Mannox' hideout. If you did everything right, your character should know the password to open it - 'truth'. Grab the sword and St. Mannox' Journal from there and go back to Caladon. Talk to all of the three people there and, when you tell everything to Hadrian he'll give you access to the three display cases behind him, containing Kraka-Tur's Eye, Finger of Mannox, and - a Glass Key.
Now, off you go the the Iron Clan entrance. Use the Glass Key on the lock and voilá! You're in. If you have Magnus in your party, you'll discover that this is his lost clan and will receive a fate point. In the next room you'll have to fight four automatons, but the reward more than makes up for it: Iron Clan Hammer, Iron Clan Platemail (the best armor in the game) and the Durin Stone, which you have to bring back to the historian in the Wheel Clan to complete the quest.
That's basically it. :D
What do you want?
- A donut.
