Rogues are the only class capable of opening locked objects (Unlike in Baldur's Gate, warriors cannot "bash" open locks, nor is there a "Knock" spell for mages). If your main character is not a rogue, your only NPC companions who can do this for you are Leliana and Zevran.
Simply select either one of them, then attempt to open the locked object. They will attempt to unlock it automatically, if they can. The lock picking ability is based on their Cunning score, so a low Cunning will lead to many locks being unpickable (even if the character has the Deft Hands skill).

It's pretty much exclusively about chests, though there are a few doors as well - but most of the locked doors in-game can only be opened with keys if at all. Anyway, to the ones you can open:
1. you must be a Rogue (..I think, though see #2).
2. cunning attribute will provide "1" point for 30, 2 for 40, 3 for 50, etc.
3. the "deft hands" field of Talents increases your ability by "1" point per talent.

The most difficult locks are I believe "5" points and result in 60 points of experience (..at least on "normal" difficulty setting).
