Honestly, I stopped here in the game, but I think I know the answer to your first question.
On the first level, the prisoner's cell requires you to pull the crank for the stretching rack in the torture chamber just above the jail (in other words: The room at the back wall, to the right of the passage with the blade pendulums (sp?).
Hope this helps.

SPOILER
The alchemist lab is in the south west portion of the map, past the temple chapel.
I am not sure about the stone door or poison door you'r talking about??
The drawbridge cannot be lowered until you make the Histachie brew(or the alchemist). You have not uncovered the whole map on the third floor yet, I take it.
Hope this helps, if you need more assistance just reply to this post?

Thx i found the crank, i was in that room a lot of times, but i dind´t see it, puff. Actually thx garluf.
Thx to you Patrick too. My problem now is that the alquemist didn´t go down, i talk to him and he says me that he will go, but he didn´t move. So i can´t get the histachi brew. I have a mage that could do the brew but i dunno how. I put in a table the mandragora herb and in the other the drakos´ tails but the southern lever don´t do anything, only appears a message telling that it´s mended, and the one of the north makes me damage. In the room with the "shreem´s" eye onto the door i try on some candles and didn´t do anything neither, it only appears a message saying that the candles are a part of the shelf. And the other two doors in the third level are closed and protected against magic and thieves. if you could help me a bit more, i´ll thank you very much.

S P O I L E R
First have you tried reading any WALKTHROUGH's for your currant position.
Don't worry about the North Lever in the chapel/temple, you have to be 9+ level cleric to operate it,(SPOILER){basicly it summons a Pit fiend in one circle and protection from evil in another}
There seems to be a glitch somehow that in the giant wyvern room there is no mandrake root. I don't know what the patch will do? I played this game twice over now, with the same party, [once in Heart of Fury}, and the second time there was no root. Very strange it was there the first time, In a rock in the giant wyvern cave. What most people do is down load DaleKeeper2.
This is a game editer. If you want it do a search in this forum for DaleKeeper2 or my thread, search 'Bard'. It is also in the sticky topic at the top.
I still do not know wich doors you speak of on the 3rd.
So have you made it past the slime room on the third floor? How about the library? There is one door all the way to the east, it will not open untill you complete all the trial or toils of the 3 floors. This is the exit to the fourth.
The alchemist makes the brew every eight hours.
The table closest to the North West cabnit is the Histachie brewing table.
Poisen brewing table is in the south east. Put all four wyvern stingers.
It glows then you have it.
The room with Sets Black Eye is only acsesable with the Modified Histahii Brew. Sounds like you may have made it here, But why you recieve that message I do not know, it should lower the bridge in the south.
I only told you stuff that was very obvious from my PRIMA's official strategy guide. I hope this helped, if not reply to this post again and we will see what we can do! Also, I know it can be complicated and tedious, just hang in there, it really is a fun game if not one of my favorites.
Good Luck
