You have to cast the spell Soul Trap on a creature then kill it while the spell is active. If you have a soul gem large enough to contain the soul, once you kill the creature, the soul will automatically become trapped in the gem.
To enchant an item, you need to have the item to be enchanted, a soul gem with a soul in it, and you need to know the spell for the effect you want to enchant the item. So if you wanted to enchant a weapon with say fire damage on strike, you need to buy the spell (NOT the scroll) for any type of fire damage. Once you have the spells, a list will show up whenever youi open the enchant dialogue with all the effect currently available to your character to use for the enchantment.
There are a couple more in-depth description at gamefaqs.com if you wanna check out the things like soul values, what gem can hold what soul, etc.

Yep. Grand soul gems are able to trap Golden Saints and Ascended Sleepers, both of which will allow you to make constant effects enchantments. In other words, the enchanted ment is always working whenever you have the item equipped, you won't need to cast it.
One note of caution though, be careful about soul trapping lower creatures like rats and such. Any soul you trap will go into the lowest rated soul gem capable of containing it. So, a rat will fit into a lesser soul gem. But, if you don't have a lesser soul gem free, the soul will be trapped in the next lowest gem. In other words, if all you have is one grand soul gem, and you soul trap a rat, you just wasted that grand soul gem with a near worthless rat soul. Try and keep a mix of soul gems ready just to be on the safe side.

Check out those pages for more on enchting points and what gems hold what.
Except for one or maybe two individuals in the game, you can't trap peoples souls.

*cough*

uhm.. i'm not suggesting this, but the early mission you get in the Balmora Mages guild, where you put a fake soul gem in the enchanter's desk while she's gone. Well, you can pick up the soul gems she accidently left lying around on her desk. One has a 300 point charge. Not that I'd know that.
