Can I load my saved game from the original Fable onto Fable TLC? Will I be missing out on anything (like quests and stuff) if I beat the game on the original Fable and then continue it on Fable TLC, or should I just start a new game on Fable TLC?

No, they're separate games. If you want to play TLC, you have to start over.
