Curio wants me to build a stronghold. I got the land deed from the guy in the vaults, but who do I get the construction permit from? The builder said Duke Dren has to provide it, but the Duke had no dialogue for me at all, except to say that Solstheim is a terible place, he's heard, but there's a boat from Khuul, if I want to go. I need that contract!

did your journal update to say that you needed to get the land deed from dren? did you talk to the guy named Driler about 'land deed' or 'stronghold' then land deed?you need to bring it up with him to trigger the duke's reaction...but even before i was hllalu i could talk to dren about more than slostheim, so i dont know whats up with your guy, you should bea ble to talk about tons of stuff, like business...

Sounds like (as JK said) something might have corrupted your file and the duke. Just out of interest are you on the PC? If you are it may well be a mod you're running, with a nolore script attached in this mod to the duke, although that only limits basic topics, he'd still talk about the stronghold deed with you because it is explictily added, but the rest of it sounds like a nolore script.
