Of course - BG will let you casually date multiple people at the same time, but once it gets serious, you monogomously decline all other romances in favor of the one you love.

If you have a multiple romance mod installed, then it will not affect the romance.
In memorian: Fiona; Ravager; Lestat; Phreddie; and all of those from the 1500 incident. Lest we forget.
