Before I start the game I was wondering about character creation. If I make a Male Gangral and a female tremere will there be a difference in the over all game? The quests and such will they change or will the game be the same with just new feats and powers?

The short answer: Kinda! The long answer: well um here goes.
As a Gangrel, you'll probably be using melee (Protean). That's FUN. My Gangrel right now is "tearing it up" if you will, running into an area, throwing on Fortitude and Protean and just going mad on everyone. My solution to problems-- kill em, or pay them if that doesn't really work. Really, its just the killing. And the claws.
As a Tremere, you'll more than likely be using Thautragry (I never spell that right, and never will) and Dominate-- you're more or less a mage. Shoot spells out at everyone, kill em just by thinking, etc-- its all very good. Lots of fun here too.
You'll notice that those two sound very similar. Well, tell the truth, they are. But that's just the general clan itself-- its up to you to customize. Do you add points to stealth, lockpicking, and hacking for your Gangrel, so that you can avoid fights or get extra goodies? Or do you add points to Persuasion/ Seduction on your Tremere, so that you can seduce or persuade everyone to do your bidding, without having to resort to violence? Or do you just go full out on your fighting skills, and let those pansies that avoid fights just rot? Do you worry about breaching the Masquerade? Are you gonna talk back to the main Prince, or be his little slave? Do you care about your humanity, or are you willing to let yourself become a real monster?
Those are the things that really make the difference. The clans just give you a general play style-- mage, fighter, etc., and then your skill choices make up the rest.
EDIT: Just reread your question and noticed I didn't answer a question. Will quests really be affected by play style? Again, the answer is kinda. As a Persuasion/ Seduction Ventrue, I was able to charm my way into everyone's love, and everyone was more willing to help me (avoiding fights, getting extra cash/xp, etc.). My Gangrel is a bit more socially backwards, though, and so has to resort to fighting quite a bit more often, and bribing when that doesn't work. Neither of them had high lockpicking/hacking/stealth, but all of those skills are very useful to use in approaching different quests.
Its been a completely different playthrough for me as my Gangrel. I would recommend playing at least twice, if not once more (just so you can play as a Toreador/ Brujah to get Celerity).

Well, save the appearances of course, the greatest differences are the discipline powers, and the clan traits. There are a few different parts in the game depending on your clan ( i.e. a Tremere character can become an apprentice at the local Tremere chantry and gets a nice heaven and an occult item if she accomplishes a quest for the Tremere Primogen ).
But in the end it all depends on your style of playing your character....
If you like Manipulation, Wits and Persuasion/Intimidation: Ventrue, Toreador
If you like Combat, Intimidation and physical prowess: Brujah, Gangrel
If you like Stealth, Researching and Information Gathering: Nosferatu
If you like Intelligence, Researching and Spells: Tremere
If you love being completely mad: Malkavian
That doesn't mean that you can't be a bad ass Toreador melee fighter for example, with Presence and Celerity as Clan Disciplines, they can be real nasty fighters.
But in the end it all depends on your style of playing your character....
