Yes, but he didn't ask about mysticism. Let's stick to the thread subject.
Destruction? A good, long distance fire spell can do a lot for you. I'm playing with the SuperAdventurers mod installed, and it adds pickpockets around Vivec. I took out one jerk running away with about 750 of my gold at 25 paces. Gotta love a nice, cheery fire.
Hah, but how do you tackle fire resistant or immune opponents?
Anyway, I also like poison because if the victim moves it looks alot like farting haha.
That doesn't make fire any less effective, because there are counters to any destructive spell--Reflect will turn them all back. And poison is too slow. An opponent with magic can remove it or heal themselves, and an opponent with healing potions can heal themselves, too, before any poison kills them. The nice thing about fire and cold-based spells is that damage can be instantaneous (if you set it up that way). It's hard to counteract a fire spell if you're already dead.

That isn't a poison spell, though, and you said poison could deliver an instakill. I'd like to know how you specifically achieve that.
Actually, the most effective destructive spell I can think of is Damage Health, because there are so few counters to it. It slips through almost all protections. Can't use it in my current game, though, since I've got a mod in place that deliberately makes it almost impossible to use. The thing's pure cheese.

You wrote,
Poison can also be instant if you set it up that way,
And I'm asking how much poison you need to make an instakill spell.

Drain Health is a very powerful spell, but it either kills or does nothing, because the victim gets his drained health back after the duration of the spell. The spell effect itself is cheap to make and easy to cast, it's the timer that makes it so expensive. The trick is to make a Drain Health 100 pts for 1 second spell: any starting character with a little skill in destruction can make it, it costs almost nothing and it instakills most monsters in the beginning of the game.

My favourite is to make a multiple effect spell (frost, fire, shock) that deilvers low levels of damage over an extended period. That way the spell does its work for you while you either pelt an opponent with arrows or hack it up with a melee weapon. It is kind of like conjuring a pet, without the hassle and high mana use.
Plus, low level, extended duration, spells often penetrate the defenses of magic resistant creatures.
Not to mention that the multiple sparks and swirls look great!
