Could you be more specific? Which door are you trying to pass?

If you are in the correct place in the sewers near Tin Can Bill then there is a gate that needs to be picked, use your Blood Buff to boost your lock picking skills.
If however you are refering to the ONLY door in the sewers near the Venture tower then you will never get that opened unless you are a Nossie as that is the Nossie Haven if I recall.
Trust me, most of the names I have been called you can't translate in any language...they're not even real words as much as a succession of violent images.

The door for the pestilence quest should be pickable. I dont' believe it required a particularly high level of lock picking as well, so use blood buff. Make sure you're in the right place; you access it through the sewer entrance which is near where Tin Can Bill was standing.

Acleacius is correct. The room near LaCroix's tower (the one with a mail box), can't be picked. It's the Nosferatu haven, and will be accessed automatically by them if they play their cards right with Gary later in the game.
