My NPC

i was wondering i never really use the npcs that i can command. for example the knights after you complete nights of the nine or the members of the dark brother hood after you become listener.

my question is, can they die, and if so do they regenerate?

Yes, your followers can die. I don't know about the Knights of the Nine but because they are unique characters I would guess that if they die they're gone forever. Mage apprentices and DB murderers will eventually be replaced by new recruits if they are killed.
Can the answer to this question be "No"?

i agree that they can die. i also think that the dark brotherhood people are the best because you can tell them to go into chamoflauge mode when they are following you. after a few of them die some thing weird begins to happen.

SPOILER!
I have had alot of them folow me and they have all died in battle... and i go to the shrine one day and get the list of people to be killed. then the next morning after i give the list to the woman there are brother hood guys show up and attack me when i go into random cities/towns. its pretty annoying but i really don't have to do anything because the guards tear them to shreds when i get there. almost like some one ordered for me to be killed...

possible spoiler

the knights of the nine also die, except for one, he is immortal during the quest but later becomes mortal. actually i lost them all in the final quest due to a bug. my screen turned messy and i could hardly find the stair to he orb. however, at the end there will be knights in the priory, without names. they can also be used as companions.
