Killing them isn't a problem. But what I would like to know is if anyone knows more about them. Their purpose and motivation are curiously absent from the lore of the game, and I've never heard any reference to them outside BG2. Are they intended merely as an easter egg of sorts--a tough battle with some good loot--or was the Twisted Rune part of a larger quest that never saw fruition? The presence of "Rune Assassins" in the tanner's basement seems to hint at some involvement with the skin dancers, but it's never elaborated upon.
The guarded compound in the Temple District raises similar questions. Tough party, good loot, secret passages that lead nowhere... if anyone has the inside scoop on either of these baddies, please share!

I believe both are part of a single quest that got scrapped. Kevin Dorner has a patch on Baldurdash that is supposed to re-activate the quest, but I have never tried it.
Also, in the Nalia romance mod, use is made of both of these, but of course this is not official.

I don't see any such patch...
Maybe we'll see more about them in BG3... (If there is one... Personally, I think I prefer party rather than individuall oriented D&D computer games)

I recall reading somewhere that the guarded compound located in the temple district is related to either Nalia's, or the fighter keeps subquest.
I think it was something to do with Roenall having Nalia imprisoned, and in order to get her off the hook, you dig up some dirt on Roenall's activites in smuggling rare/valuable gems/jewels..... a bit vague I know, but I have a bad memory at the best of times, let alone trying to remember a tidbit of info from a year or two ago

I think you're confusing the Delryn rival family in the Bridge district (the surname escapes me) - part of Anomen's test of knighthood, with the twisted rune, who are an entirely separate entity.
The slavers in the temple district guarded compound - also completely unrelated to the other two - are supposedly connected to the slavers in the slums, but I'm not aware of any practical implications.

Officer Dirth in the Sea Bounty does tell you how to find evidence against Roenall and save Nalia. However, I do not believe the Nalia and Temple Slaver quests are otherwise related.
I've seen debates over this around here before and other places on the net. Here's my two cents:
Regarding the compound in the temple district: I thought I had read elsewhere this was supposed to be the final tie in to the slavers quest originating at the copper coronet but it was never completed.
Why? Perhaps because the fight is a bit tough for characters of an introductory level to the game. Not that it can't be won, but if one were new to the game, that fight would pose quite a challenge. It's also a bit early in the game to be rewarded with the celestial fury. That weapon makes a huge difference, and maybe the developers didn't want to see players running around with it all game. Since most people do the slavers quest pretty early in the game, had they tied in the guarded compound to the quest, everyone would be running around with celestial fury for most of the game.
Regarding the twisted rune: That quest is triggered if one selects the quest option when casting a limited wish spell. However, once you know where it is, you can use limited wish spells for other things.
Crush enemies, see them driven before you, and hear the lamentations of the women.
maybe you're right @galuf. All I know is first time through the game, I found something mentioning the twisted rune quest and the use of the rogue stone.

There's probably a good article in all the features and localities that were left out of popular games due to time constraints. Might and Magic VI, for instance, was supposed to feature a rumor system that could pass along ideas of quests from a great distance, but with increasing distortion. It's fun to read the original interviews and press releases of a product, then look at the finished code to see what was left out.
